چکیده

Twenty-two patients with childhood ALL were analyzed for the configuration of their Ig (innunoglobulin) and TcR-6 (T cell receptor delta) genes by Southern blot analysisThe detection ofgene rearrangenents indicatedwhether IgH (itnmunoglobulin heavy chain) or TcR-6 junctional regiotts should be analyzed by PCR (Polymerase chain reaction). The nucleotide sequence ofjunctional regions of the rearranged IgH and TcR-ô genes are deternined by direct sequencing of PCR products of these regiotrs. Based on the sequence data, patient-specific oligonucleotide probes were designedSubsequently, bone narrow and/ or peripheral blood sanples taken daring andafier treattnent, will be analyzedwiththe PCRtechnique, using patient specific oligonucleotide probesfor the detection ofleukenic cells. Five patient specffic probes have been designed and tested in specificity and sensitivity.

The relation between end of induction minimal residual disease and different risk factors in patients with acute lymphoblastic leukemia

Background: Malignant disorder with B or T stem cell basis leads to development and continuation of acute lymphoblastic leukemia (ALL) due to aggregation of blast cells in bone marrow. The environmental, genetic, and demographic factors may influence the disease relapse. The objective of this study was to assess the relation between end of induction minimal residual disease and different risk f...

High applicability of ASO-RQPCR for detection of minimal residual disease in multiple myeloma by entirely patient-specific primers/probes

Allele-specific oligonucleotide real-time quantitative PCR (ASO-RQPCR) is a standardized technique for detection and monitoring of minimal residual disease (MRD) in acute lymphoblastic leukemia (ALL) but not multiple myeloma (MM) due to a low applicability inherent with presence of somatic hypermutation. Herein, by a staged PCR approach and sequencing, clonality and tumor-specific complementari...

Detection of minimal residual disease in T-cell acute lymphoblastic leukemia using polymerase chain reaction predicts impending relapse.

After achieving remission, approximately one-third of patients with T-cell acute lymphoblastic leukemia (T-ALL) relapse due to the resurgence of residual leukemic cells that cannot be detected in remission by morphologic methods. Thus, the early detection of residual disease is highly desirable to monitor the efficacy of therapy, or to institute an alternative mode of therapy. Toward this aim, ...
